中文摘要:为明确高寒草地牧草矮生嵩草(Kobresia humilis)内生细菌262AG6的生防潜力,采用平皿对峙法测定其抑菌功能,并综合形态学特征和16S rDNA序列分析对其进行鉴定。结果表明,262AG6对马铃薯炭疽病菌(Colletotrichum coccodes)的抑制效果达到81.33%,对其他多种病原真菌也具有良好抑制能力,且抑菌能力稳定;在含100 mg·L-1色氨酸和不含色氨酸的King培养液中分泌IAA的量分别为8.04 mg·L-1 和7.98 mg·L-1;具有溶解无机磷和固氮的能力;262AG6革兰氏染色呈阳性,杆状,菌体大小为( 1.63~ 3.65) μm× (0.38~0.95) μm,综合形态特性、16S rDNA和gyrB DNA序列分析将菌株262AG6鉴定为解淀粉芽胞杆菌(Bacillus amyloliquefaciens)。

Identification and Determination of Antagonistic Function of Endophytic Bacteria 262AG6 from Kobresia humilis in Alpine Grassland
Abstract:In order to evaluate the biological control potential of endophytic bacteria 262AG6 isolated from Kobresia humilis at alpine grassland, the inhibitory effects were studied by dual culture against Colletotrichum coccodes, and the 262AG6 was identified by morphological characteristics and 16S rDNA sequence analysis.The results showed that the inhibition ratio of 262AG6 strain against Colletotrichum coccodes was as high as 81.33%, and also presented a higher stability and inhibitory effects on other species of pathogens.The secreted contents of IAA were 8.04 mg·L-1 with 100 mg· L-1 tryptophan on the King media and 7.98 mg·L-1 without ryptophan, and the strain also had ability to dissolve inorganic phosphorus and to fix nitrogen.The thalli was rod shaped, gram positive and (1.63-3.65) μm ×(0.38-0.95) μm in size.The 262AG6 was identified as Bacillus amyloliquefaciens based on 16S rDNA and gyrB DNA sequence analysis and the morphological characteristics.
