地钱部分MYB家族基因在不同发育时期、部位以及激素处理下表达量差异分析

中文摘要:为了研究植物适应陆地生活的机制,以较原始的陆生植物地钱(Marchantia polymorpha L.)作为研究对象,分析其部分MYB(v-Myb avian myeloblastosis viral oncogene homolog)家族基因在不同发育时期,不同部位以及不同激素处理下的表达量变化。构建系统进化树确认地钱与拟南芥(Arabidopsis thaliana)中MYB转录因子亚家族S14、S28和 AtCDC5 同源的基因,使用qRT-PCR对不同生长时期、组织以及激素处理下目的基因表达量进行分析。qRT-PCR分析结果表明, MpR2R3-MYB20在胞芽中表达量最高,胞芽杯中最低,且在0.1 μmol/L IAA诱导下表达量显著上调; MpR2R3-MYB8在分生区高表达,激素处理下表达量变化较小; MpR2R3-MYB3 MpCDC5在3周分生区高表达,IAA处理后下调; MpR2R3-MYB18主要在胞芽和胞芽杯中表达,且受ABA诱导大幅上调; MpR2R3-MYB16在全株中都有较高表达,在0.1 μmol/L IAA处理下显著上调,在其他激素处理下显著下调。这些结果说明上述基因在地钱不同器官或组织中具有调节生长、发育和抗逆的重要功能。

Analysis of Expression Pattern of Some Liverwort MYB Genes at Various Developmental Stages,Different Tissues and Their Responses to Plant Hormone Treatments
Abstract:To elucidate the mechanisms that enable plants to adapt to a terrestrial environment,this study determined the expression level of some MYB genes in Marchantia polymorphaof a primitive land plant in different tissues at various developmental stages and after the plants were treated with several plant hormones. A phylogenetic tree was constructed to identify liverwort homologs to Arabidopsis thaliana MYB transcription factors in the S14,S28 subfamilies and AtCDC5. The expression level of these genes in different tissues and developmental stages,before and after treatment with auxin and ABA,were compared by using real-time quantitative PCR. MpR2R3-MYB20 had the highest and lowest expression in gemma and gemma cup,respectively,and it was significantly up-regulated under 0.1 μmol/L IAA. MpR2R3-MYB8 was expressed highly in meristem zone,which had little change under hormone treatments. MpR2R3-MYB3 and MpCDC5 were highly expressed in the meristem,but down-regulated under IAA treatment. MpR2R3-MYB18 was mainly expressed in gemma and gemma cup and up-regulated by ABA. MpR2R3-MYB16 detectable in the whole plant showed a high level of expression under 0.1 μmol/L IAA treatment,but it was at lower level under other treatments.These results suggested that the above-described genes had important roles in the growth,development and stress responses in different organs or tissues.
