运输应激对马体温、血液指标及HPTNF-αIFN-γ mRNA表达水平的影响

中文摘要:旨在研究运输应激对马体温、血常规指标及HPTNF-αIFN-γ mRNA表达水平的影响,测定马运输前1 h、运输中7 h及运输后1 h和9 h的体温及血液指标,RT-qPCR方法检测外周血淋巴细胞中HPTNF-αIFN-γ的mRNA表达水平。结果显示,与运输前1 h相比,运输中7 h时马体温显著增高,运输后1 h的体温极显著增高,运输后9 h的体温逐渐下降但仍显著高于运输前;运输后9 h红细胞平均体积(MCV)显著降低,其他指标无明显差异;外周血淋巴细胞中HPTNF-αIFN-γ的mRNA表达量均呈上升趋势,HPIFN-γ的mRNA表达量在运输后1 h达到高峰,且与运输前相比差异极显著,运输后9 h逐渐恢复;TNF-α的mRNA表达量一直呈上升趋势,运输后9 h TNF-α的mRNA表达量极显著升高。研究表明运输应激会导致马体温上升,MCV下降,上调马外周血淋巴细胞中HPTNF-αIFN-γ的mRNA表达水平,结果为进一步阐述运输应激的致病机制提供依据。

Effects of Transport Stress on Body Temperature,Blood Indexes,mRNA Levels of HP,TNF-α and IFN-γ in Horse
Abstract:To investigate the effect of transport stress on body temperature,blood indexes,HP,TNF-α and IFN-γ of horses,we measured the body temperature and blood indicators of horse at 1 h before transportation,7 h during transportation,1 h and 9 h after transportation,and we also detected the expression levels of HP,TNF-α and IFN-γ mRNA in peripheral blood lymphocytes by fluorescent quantitative PCR. The results showed that compared with 1 h before transportation,the body temperature increased significantly at 7 h after transportation,and at 1 h after transportation. The body temperature of 9 h after transportation gradually decreased,but it was still significantly higher than 1 h before transportation;the mean erythrocyte volume (MCV) of horse after 9 h transportation significantly reduced,while other indexes showed no significant difference.The mRNA levels of HP,TNF-α and IFN-γ in peripheral blood lymphocytes showed an increasing trend. The expression levels of HP and IFN-γ peaked at 1 h after transportation and it had significant difference,and recovered gradually at 9 h after transportation. TNF-α expression level kept increasing and was significantly higher at 9 h after transportation. In summary,transport stress in horsescan cause the increase of body temperature,decreaseof MCV level,and up-regulation of gene expressions in HP,TNF-α and IFN-γ of peripheral blood lymphocytes. These results provide a basis for further elucidating the pathogenesis of transport stress.
