葡萄NF-Y核因子家族的鉴定与表达分析

中文摘要:为阐明葡萄NF-Y核因子的理化性质、进化关系和在非生物胁迫下的表达情况,本研究从葡萄全基因组中鉴定出24个VvNF-Y核因子家族成员。生物信息学分析表明其编码的氨基酸数目为132~345 aa,pI为4.62~9.83,蛋白二级结构以α-螺旋和不规则卷曲为主,亚细胞定位发现葡萄NF-Y基因主要定位于细胞核中;系统进化分析表明进化树可分为3个亚族;顺式作用元件分析发现葡萄NF-Y基因家族具有大量核心元件,如光响应元件和响应多种激素和环境信号的元件,且多序列比对分析表明VvNF-Y的3个亚族均含有保守区;此外,选择性压力分析表明8对基因对的Ka/Ks均小于0.5,在进化过程中受到纯化选择作用;qRT-PCR结果表明在500 μmol·L-1 ABA、10% PEG和200 mmol·L-1NaCl处理时, VvNF-YA2VvNF-YB3VvNF-YB4VvNF-YC3的表达量显著上升,且在10% PEG条件下, VvNF-YA1的表达量也显著上升,其余NF-Y核因子家族成员在受到ABA、NaCl和PEG处理时表达量下降。试验结果表明葡萄NF-Y核因子在参与葡萄不同的非生物胁迫时可能发挥不同的作用,为葡萄抗逆性研究提供了候选基因。

Identification and Expression Analysis of NF-Y Nuclear Factor Family in Grape
Abstract:To clarify physical and chemical properties and evolutionary relationships of grape NF-Y nuclear factors and their expressions under abiotic stress, 24 family members of VvNF-Y nuclear factors were identified from grape widen-genome. The analysis of bioinformatics showed that the number of the encoded amino acids ranging from 132 to 345 aa and Isoelectric Point (pI) of these amino acids was between 4.62 and 9.83, and their protein secondary structures were mainly α-helix and random coil and the subcellular localization indicated that NF-Y gene family was mainly expressed in the nucleus. The phylogenetic analysis revealed that the NF-Y gene family members could be divided into three main subgroups. The cis-acting element analysis found that the grape NF-Y gene family included a large number of core elements, such as light responsive element, multi-hormones and environmental signals responsive elements.The multiple sequence alignment analysis indicated that there wasa conserved domain in all three subgroups of NF-Y. In addition, the selective pressure analysis showed that Ka/Ks of all eight gene pairs were less than 0.5, and NF-Y genes were subject to purification selection in the process of evolution. Quantitative Real-time PCR (qRT-PCR) analysis showed that the expression levels of VvNF-YA2, VvNF-YB3, VvNF-YB4 and VvNF-YC3 increased considerably after being treated with 500 μmol·L-1 ABA, 10% PEG and 200 mmol·L-1 NaCl respectively. The expression level of VvNF-YA1 also increased significantly under 10% PEG treatment, while the level of other NF-Y nuclear factor members obviously decreased under ABA, NaCl and PEG treatments. The results of this study indicated that the nuclear factors of VvNF-Y may play different roles in participating in different abiotic stress in grape, which provided candidate genes for research in grape stress resistance.
