中文摘要:为明确陕西省西洋参根结线虫病病原,采用常规形态鉴定结合分子生物学方法,对在陕西省留坝县严重发生的西洋参根结线虫病进行病原鉴定。结果表明:二龄幼虫蠕虫形,口针基部球小而圆,尾部透明区明显,尾尖钝圆;雌虫虫体梨形或近椭圆形,口针杆部后部稍宽,口针基部球近圆形较小有缢缩。雌虫会阴花纹呈近圆形,线纹平滑呈波浪状,尾端区有刻点,背弓较为低平。结合特异性PCR扩增及根结线虫属rDNA-ITS序列系统发育树构建结果,该病原鉴定为北方根结线虫Meloidogyne hapla。 |

Pathogen Identification of Panax quinquefolium Root-knot Nematode Disease in Shaanxi of China |

Abstract:In order to clarify the pathogen species of Panax quinquefolium root-knot nematode disease in Shaanxi ,the root samples with obvious root-knots of Panax quinquefolium were collected for isolating root-knot nematodes from P.quinquefolium planting area in Liuba County,Shaanxi Province.The pathogenic nematodes were identified by observing morphological characteristics of females,second-stage juveniles and perineal pattern,sequence alignments,phylogenetic analysis and specific PCR amplification.The results showed that the pathogen species of P.quinquefolium root-knot nematode disease in Shaanxi was M.hapla. |
