外源褪黑素对NaCl胁迫下千屈菜幼苗生长及抗性生理的影响

中文摘要:为研究外源褪黑素对NaCl胁迫下千屈菜幼苗生长及抗性生理的影响,以千屈菜种子为材料,选择对千屈菜幼苗生长发育、光合色素影响显著的10 g·L-1 NaCl溶液作为盐胁迫处理,然后使用不同浓度(0、50、100、150、200 μmol·L-1)的外源褪黑素喷施千屈菜幼苗,检测外源褪黑素对NaCl胁迫下千屈菜幼苗的主要生长指标和生理指标的影响。结果表明,NaCl胁迫显著抑制千屈菜幼苗的根长、株高、鲜质量等,还导致千屈菜幼苗丙二醛(MDA)、脯氨酸和可溶性蛋白含量显著高于蒸馏水处理的对照组(CK),而超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)等抗氧化酶活性和叶绿素含量显著低于对照。与单独NaCl胁迫处理(MT0)相比,低浓度的褪黑素能在一定程度上缓解NaCl胁迫对幼苗造成的伤害,而高浓度褪黑素则会使伤害加剧,100 μmol·L-1褪黑素处理的千屈菜幼苗生长得到显著改善,幼苗根长、株高、鲜质量与单独NaCl胁迫处理(MT0)比较分别升高25.61%、38.15%和53.01%,干鲜比降低62.75%,脯氨酸和可溶性蛋白含量分别增加30.55%和35.86%,叶绿素a、叶绿素b和总叶绿素含量分别提升34.46%、34.16%、 34.39%。SOD、POD、CAT酶活性分别上升68.91%、30.61%和19.55%,MDA含量较单独NaCl胁迫处理下降30.67%。外源褪黑素浓度在低于150 μmol·L-1时能够减轻盐胁迫带来的伤害,超过150 μmol·L-1时会使伤害加剧,缓解千屈菜幼苗NaCl胁迫伤害效果最佳的褪黑素浓度为100 μmol·L-1

Effect of Exogenous Melatonin on Growth and Resistance Physiology of Lythrum salicaria Seedlings under NaCl Stress
Abstract:This study investigates the effects of exogenous melatonin on growth and resistance physiology of Lythrum salicaria seedlings under NaCl stress. Lythrum salicaria seedlings were used as materials,a 10 g·L-1 NaCl solution,which has significant effects on the growth,development,and photosynthetic pigments of seedlings,was chosen as salt stress treatment,and then Lythrum salicaria seedlings were sprayed with exogenous melatonin at different concentrations. An experiment was conducted to analyze the effect of exogenous melatonin on the main growth indexes and physiological indexes of Lythrum salicaria seedlings under NaCl stress. The results showed that NaCl stress significantly inhibited root length,plant height and fresh and dry mass of Lythrum salicaria. Malondialdehyde,proline and soluble protein contents of Lythrum salicaria seedlings were significantly higher than those in CK control group,while the activities of antioxidant enzymes such as superoxide dismutase (SOD),peroxidase (POD) and catalase (CAT) of Lythrum salicaria seedlings were significantly lower than those in CK control group. Chlorophyll content was significantly lower than that of the control when compared with NaCl stress alone (MT0).A low concentration of melatonin alleviated the damage to seedlings caused by NaCl stress to a certain extent,while high concentration of melatonin aggravated the damage. The growth of Lythrum salicaria seedlings treated with 100 μmol·L-1 melatonin was significantly improved. The root length,plant height,fresh mass and dry mass of seedlings increased by 25.61%,38.15% and 53.01%,respectively,compared with MT0 treated with NaCl alone,the contents of proline and soluble protein increased by 30.55% and 35.86%,and the contents of chlorophyll a,chlorophyll b and total chlorophyll increased by 34.46%,34.16% and 34.39%,respectively. The activities of SOD,POD and CAT increased by 68.91%,30.61% and 19.55%,respectively,and the content of malondialdehyde decreased by 30.67% compared with NaCl stress alone. Exogenous melatonin concentration can alleviate the damage caused by NaCl stress when it is lower than 150 μmol·L-1,and aggravate the damage when it is higher than 150 μmol·L-1. The most suitable melatonin concentration for the growth of Lythrum salicaria seedlings under NaCl stress is 100 μmol·L-1.
