犬源细小病毒兰州株的分离鉴定与VP2基因的遗传进化分析

中文摘要:为了解近年来甘肃省兰州市犬细小病毒(Canine parvovirus,CPV)的基因型分布情况及遗传进化方向,于兰州市宠物医院采集8份细小病毒阳性(试纸条检测)的犬粪便样品,使用F81细胞分离病毒;利用PCR、间接免疫荧光试验、血凝试验鉴定所分离的病毒;随后分析病毒基因型和 VP2基因的遗传进化关系。结果显示,共分离得到7株细小病毒,1株为New CPV-2a型CPV,5株为CPV-2c型CPV,1株为猫细小病毒(Feline parvovirus,FPV)。VP2氨基酸序列同源性及进化分析结果显示,分离到的CPV-2c型毒株VP2同源性为100%,与北京、江苏等地分离株的亲缘关系较近;New CPV-2a型毒株与吉林、西安等地的分离株亲缘关系较近;犬源猫细小病毒(LZ05)与已报道的FPV毒株相比,存在91S→A、322V→T独特的突变,且与2020年北京一株犬源猫细小病毒分离株亲缘关系最近。

Isolation,Identification of Parvovirus from Canine in Lanzhou and Phylogeny Analysis of VP2 Gene Sequences
Abstract:To investigate the genotype distribution and genetic evolution of canine parvovirus (CPV) in Lanzhou in recent years, eight fecal samples tested positive using a canine parvovirus rapid test kit were collected from pet hospitals in Lanzhou. F81 cells were used for isolating parvovirus strains; PCR,indirect immunofluorescence and hemagglutination assays were conducted to identify the isolated strains; additionally,the genotype and genetic evolution relationship were analyzed. The results showed that seven parvovirus strains were isolated,one strain belonged to the New CPV-2a type,five strains were classified as CPV-2c type,and one strain was identified as Feline parvovirus (FPV). The homology of amino acid sequences and evolutionary analysis showed that the VP2 sequences of these CPV-2c strain was 100% homologous and closely related to the strains found in Beijing and Jiangsu. The New CPV-2a strain exhibited similarities to those found in Jilin and Xi’an. Compared with the reported strains,FPV of canine origin (LZ05) had unique mutations (91S→A and 322V→T),and was found to be most closely related to a strain isolated in Beijing in 2020.
