中文摘要:为了提升亚麻荠的产量及含油量,本研究通过农杆菌侵染转化法,将油菜中DGAT基因的4个拷贝分别转入亚麻荠中,得到亚麻荠过表达材料。与野生型种子相比,转基因系种子更加圆润饱满,更平滑有光泽感,种子体积变大,同时种子的质量也增加。种子含油量测定结果显示,过表达系BnaDGAT1-1 OE、BnaDGAT1-2 OE、BnaDGAT1-3 OE和BnaDGAT1-4 OE种子含油量均高于野生型,其中BnaDGAT1-3 OE含油量最高。通过qRT-PCR测定与油脂相关基因的表达量,结果显示BnaDGAT1-1 OE、BnaDGAT1-2 OE、BnaDGAT1-3 OE和BnaDGAT1-4 OE的CsFAE1CsFATACsFATB的表达量均高于野生型,而CsSAD则不变。

Effects of Heterologous Expression of DGAT1 Gene on the Expression of Genes Related to Oil Content and Oil Synthesis Incamelina Seeds
Abstract:In this study,four homologous of Brassica napus DGAT1 (BnaDGAT1) were cloned and transferred into Camelina sativa through the floral dip method to improve the yield and oil content of camelina.The overexpression lines of BnaDGAT1 in camelina were obtained.Compared with the wild-type seeds,the seeds physiology of transgenic lines were more rounded and full,smoother and shiny.The volume and weight of transgenic seeds were also increased significantly.These results showed that the BnaDGAT1-1 OE,BnaDGAT1-2 OE,BnaDGAT1-3 OE and BnaDGAT1-4 OE overexpression lines comprised higher seed oil content than the wild type,while BnaDGAT1-3 OE overexpression lines exhibit the maximum oil content.The expression level of lipid-related genes was determined by qRT-PCR,the results indicated that the expression level of CsFAE1CsFATACsFATB in BnaDGAT1-1 OE,BnaDGAT1-2 OE,BnaDGAT1-3 OE and BnaDGAT1-4 OE overexpression lines were significantly increased as compared to that of wild-type,while the expression level of CsSAD remained unchanged.This study reveals a theoretical basis for further exploring the improvement of the oil content of Camelina sativa and the related pathways of oil biosynthesis in Camelina sativa.
