| 中文摘要:GA20氧化酶是赤霉素生物合成途径中的重要氧化酶,在植物株高调控中发挥着重要作用。为研究 TeGA20ox在万寿菊中的生物功能,本文以万寿菊矮杆品系‘c-16’、中杆品系‘72’、高杆品系‘2-2’为试验材料,采用同源克隆结合RACE技术获得 TeGA20ox基因全长,对其进行生物信息学分析、亚细胞定位,并用qRT-PCR技术检测 TeGA20ox基因在万寿菊不同时期以及不同组织中的表达量。结果表明,克隆到万寿菊 TeGA20ox基因全长为1 550 bp,CDS长度1 149 bp,共编码382个氨基酸。TeGA20ox蛋白分子质量43.55 ku,为亲水蛋白且无跨膜结构,TeGA20ox蛋白与菊薯、向日葵、微甘菊亲缘关系较近。并构建表达载体瞬时转化烟草,该蛋白定位于细胞膜和内质网上。‘72’和‘2-2’的表达量随着株高的增高逐渐升高,呈正相关趋势。 TeGA20ox在万寿菊不同组织中的表达存在差异,矮杆品系‘c-16’中 TeGA20ox的最高表达部位为幼嫩叶片,而中杆品系‘72’和高杆品系‘2-2’中茎表达量最高。以上结果表明 TeGA20ox基因的表达与万寿菊株高存在一定关系, TeGA20ox在万寿菊株高调控方面起到了重要作用。 |

| Cloning,Subcellular Localization and Expression Analysis of the Plant Height-related Gene TeGA20ox in Tagetes erecta |

| Abstract:GA20-oxidase (GA20ox) is a key enzyme in the gibberellin biosynthetic pathway and plays an important role in regulating plant height.To investigate the biological function of TeGA20ox in marigold (Tagetes erecta),the full-length gene was isolated using homologous cloning combined with RACE technology,with dwarf line ‘c-16’,medium line ‘72’,and tall line ‘2-2’ as experimental materials.Bioinformatics analysis and subcellular localization of the TeGA20ox gene were performed,and the expression patterns in different developmental stages and tissues were analyzed using qRT-PCR.The results showed that the full length of TeGA20ox gene was 1 550 bp,with the CDS of 1 149 bp encoding 382 amino acids.The molecular mass of the TeGA20ox protein was 43.55 ku,which is a hydrophilic protein without transmembrane domains.Phylogenetic analysis revealed that TeGA20ox protein closely related to its homologs in Smallanthus sonchifolius,Helianthus annuus and Mikania micrantha.The expression vector was constructed and transiently expressed in Nicotiana benthamiana,and the protein was localized to the cell membrane and endoplasmic reticulum.The TeGA20ox expression in lines ‘72’ and ‘2-2’ gradually increased with plant height,suggesting a positive correlation.The expression also showed tissue-specificity.In the dwarf line ‘c-16’,the highest expression was observed in young leaves,whereas in the middle line ‘72’ and tall line ‘2-2’,stems showed the highest expression level.These results indicate that the expression of TeGA20ox gene is closely associated with plant height in marigold,and suggest that TeGA20ox plays an important role in regulating marigold plant stature. |
