薰衣草内生真菌分离、筛选与菌株Alternaria alternata XYC-7次级代谢产物鉴定及抗氧化活性分析

中文摘要:本研究旨在从薰衣草中筛选兼具抑菌与抗氧化活性的内生真菌资源、并鉴定其活性代谢产物。采用组织块法分离薰衣草根、茎、叶的内生真菌,通过形态学特征与ITS rDNA序列分析对菌株进行鉴定;利用琼脂块法和滤纸片法测定菌株对革兰氏阳性菌(金黄色葡萄球菌、枯草芽孢杆菌)及革兰氏阴性菌(大肠杆菌)的抑菌活性,并通过DPPH自由基清除试验评估抗氧化能力。结果表明:从薰衣草中分离的13株内生真菌中,发现其中有10株内生真菌对供试菌显示出抑菌活性,其中菌株XYC-7的菌株抑菌效果最强,对大肠杆菌、金黄色葡萄球菌和枯草芽孢杆菌的抑菌圈直径分别达2.6 cm、2.5 cm和2.7 cm,表现出广谱抑菌性。该菌株发酵液的乙酸乙酯萃取物在浓度1 mg/mL时,对DPPH自由基清除率为89.14%,显示出显著的抗氧化潜力。进一步通过高效液相色谱(HPLC)分析,发现其活性成分保留时间与花色苷标准品一致,含量为0.826 mg/L,表明黄酮类物质(花色苷)是关键的活性成分。结合形态学与分子生物学鉴定,最终确认XYC-7为互生链格孢菌(Alternaria alternate)。

Isolation and Screening of Endophytic Fungi from Lavender and Identification of Secondary Metabolites with Antioxidant Activity from Strain Alternaria alternata XYC-7
Abstract:Endophytic fungi from Lavandula angustifolia Mill. were isolated and screened for antibacterial and antioxidant activities,followed by identification of active metabolites. Fungi were isolated from the roots,stems,and leaves using tissue culture and identified based on morphological characteristics and ITS rDNA sequencing. Antibacterial activity against Staphylococcus aureus,Bacillus subtilis(Gram-positive),and Escherichia coli (Gram-negative) was evaluated using agar block and filter paper disc methods. Antioxidant capacity was determined via DPPH radical scavenging. Among 13 isolated strains,10 exhibited antibacterial activity. Strain XYC-7 demonstrated potent,broad-spectrum activity,with inhibition zones of 2.6 (E. coli),2.5 (S. aureus),and 2.7 (B. subtilis) cm. Its ethyl acetate extract showed significant antioxidant activity (89.14% DPPH scavenging at 1 mg/mL). HPLC analysis identified a compound co-eluting with an anthocyanin standard (retention time match),quantified at 0.826 mg/L,suggesting anthocyanins (flavonoids) as key bioactives. Integrated morphological and molecular characterization identified strain XYC-7 as Alternaria alternata. These findings suggest that A. alternate XYC-7 is a promising source of dual-activity metabolites.
